To get the Stupefy spell you have to progress through the quest The Path to Hogwarts. This is the first main quest you will get right after creating your character, and you will learn a bunch of useful spells besides Stupefy along the way. Now that you have the Stupefy spell, you can use it in battle, but if you want to increase its effectiveness, it is best to use it with Protego. You can unlock Protego during the same quest, so you can try them both together.
Protego is a shield spell that will protect you, and right after using Protego, you can counter oncoming attacks with Stupefy automatically. This will stun the enemy, giving you a split second to cast any attacking spell. Any enemy stunned will receive increased damage from spells. You can quickly swap targets to aim and shoot the spell as well.
