You have to complete the main story quest Crisis at Hyrule Castle to unlock the Paraglider. You will bump into this quest as you play the game, and you will encounter it very early on. The first objective is to find and talk to Captain Hoz in Hyrule Castle, then visit Purah at Lookout Landing. You can then visit the Skyview Towers to get the Paraglider as a gift.
To use the Glider, all you have to do is jump off a high point and press X to whip out the glider. You will automatically de-equip it when you land. If you wish to know how many quests are left till you reach the point of getting the glider, you can always check your Adventure Log to see how far you need to progress.
