How to Find Barkenstein in Tiny Tina’s Wonderlands
When you complete the main story mission in Tiny Tina’s Wonderlands, you will come across the Chaos Chamber which has plenty of monsters as well as bosses for you to beat, and high rewards if you manage to clear the levels. There are four raid bosses that are particularly lethal, and one of them is known as Barkenstein.

Barkenstein is a boss you will find within the halls of the Chaos Chamber who will attack you with poison. Make sure you are prepared for the boss level fight before you reach his dungeon. You can unlock his battle by completing a puzzle within the Chaos Chamber which will summon him through a red portal, for which there are quite a few steps you need to follow.
To get to the puzzle, you will need to find a green run in your random Chaos Chamber dungeons. Once you find it, you can interact with it for the green orb to fly across the room and hit the ground. It will spawn a mushroom which you need to jump on, and shoot at the balls that form in the air. This will occur three times, after which you will have completed the puzzle. You can now move onward to fighting Barkenstein by using the red portal which will lead you to them.
