The door can be unlocked a bit later into the game, after reaching the Four Belfries Site of Grace. You can get here in the western area of Liurnia of the Lakes. You will need to get to the fourth Belfry at the peak of the hill and get to the chest. Open it to get Imbued Sword Key. This will open up any of the waygates in the other Belfry locations. Head over to the second Belfry to the top east of the hill that has a waygate that will lead you to the Chapel of Anticipation. This will lead you to the place where you started the game and also meet with the Grafted Scion. Upon defeating it, go up the staircase and check out the door on the right. This is the door that was locked at the start. Go inside and explore the area to get some items like the Stormhawk King Ash of War and the Stormhawk Deenh Spirit Ash.
